چکیده

Anisotropy of anionic sites on plasma membranes has been implicated in contact phenomena at cell surfaces (16, 17). Recent evidence indicates that charge anisotropy may be important among intracellular membranes also, since distribution of anionic sites has been related to structural organization of mitochondrial membranes (8). The use of cationic ferritin has been introduced by Danon et al. (2) as a marker to demonstrate anionic sites on membrane surfaces by electron microscopy. This visual probe has been applied to study anionic sites on cell surfaces (2, 7) and on mitochondrial membranes (8). In the present study, cationic ferritin has been used to demonstrate anionic sites on the surface of the Golgi complex in relation to the structural organization of this membrane system.

Analytical study of microsomes and isolated subcellular membranes from rat liver VIII. Subfractionation of preparations enriched with plasma membranes, outer mitochondrial membranes, or Golgi complex membranes

Preparations enriched with plasmalemmal, outer mitochondrial, or Golgi complex membranes from rat liver were subfractionated by isopycnic centrifugation, without or after treatment with digitonin, to establish the subcellular distribution of a variety of enzymes. The typical plasmalemmal enzymes 5'-nucleotidase, alkaline phosphodiesterase I, and alkaline phosphatase were markedly shifted by dig...

Differential distribution of xyloglucan glycosyl transferases in pea Golgi dictyosomes and secretory vesicles

Rate-zonal centrifugation of pea (Pisum sativum var. Alaska) stem microsomal membranes on a linear Renografln gradient separated Golgi secretory vesicles from dictyosomes. Secretory vesicles possessed high levels of xyloglucan fucosyl transferase activity, which effects the final decoration of stem xyloglucan side-chains, but lacked substantial xyloglucan xylosyl transferase activity, which is ...

Interaction of lectins with proteins of the endoplasmic reticulum and Golgi system of rat liver.

The interaction of glycoproteins of rough and smooth microsomal and Golgi membranes with Sepharose-bound lectins has been studied. One of these lectins was a crude preparation from wheat germ lipase which was found to bind primarily to N-acetyl neuraminic acid. Rough microsomes, smooth microsomes and Golgi membranes contain glycoproteins which bind to Concanavalin A (Con A specific for mannose ...

Golgi Cisternal Unstacking Stimulates COPI Vesicle Budding and Protein Transport

The Golgi apparatus in mammalian cells is composed of flattened cisternae that are densely packed to form stacks. We have used the Golgi stacking protein GRASP65 as a tool to modify the stacking state of Golgi cisternae. We established an assay to measure protein transport to the cell surface in post-mitotic cells in which the Golgi was unstacked. Cells with an unstacked Golgi showed a higher t...

Membrane differentiation in the Golgi complex of Micrasterias denticulata Bréb. visualized by freeze-etching.

INTRODUCTION In recent years the polarity of dictyosomes has been well established, both iu functional and in morphological terms (Mollenhauer & Morre, 1966; Favard, 1969). Although many of the observed differentiations across stacks of dictyosome cisternae probably result from changes in the Golgi membranes and their associated enzymes, it has only recently been possible to obtain electron-mic...
